My interest is in all business-strike coins minted 1900 and later in circulated condition, silver dollars through cents.
I get my joy from finding the coins - not in simply purchasing them. So, every week, I go to my bank and pick up boxes to search through. I keep the ones I want and take the rest back to a different bank. I am somewhat fortunate in that I have friends working at several banks, with the closest being less than a mile away with a coin sorter in the lobby.
I am currently working on filling two Dansco albums - Lincoln Memorial Cents and Jefferson Nickels. Right now I search a box of cents and a box of nickels a week. Very shortly I plan on adding a box of halves a week as well.
Once I have all the coins from 1900 till now (if that ever happens) - then I will start on commemoratives, again focusing on those minted 1900 and later.

I keep all wheat cents, foreign coins, copper cents 1959 - 1981, and all coins containing silver.
A note about errors and varieties - I look for them, but don't really consider them to be part of my collection. So, doubled dies, repunched mintmarks, close and wide AM - they're all nice, but I don't have to have them. I do, however, want the large and small date varieties - for some reason, I look at these a little differently.
